you can do it like this like this:
http://code.google.com/apis/maps/documentation/javascript/v2/examples/directions-advanced.html

to get driving or walking directions in v2 or driving, walking and
bicycling (US only) directions in v3, but neither API's documentation
says anything about retrieving public transport directions.
