I'm very new to this Phyton and i just downloaded django package. What i wanted to ask was,..can i make a social site where the users itself can customize their themes or the layout???

Welcome to Python and Django! Of course you can. I'd suggest starting with the Django Tutorial to understand the framework and of course learning a bit about the Python programming language before jumping into a task that large.

For an example site that provides this functionality (still a work in progress), check out fireflie.com. You have to register to see the functionality but feel free to ignore the subscription after 8 days -- unless you're a musician and want to actually use the site.

(Disclaimer: I am the lead developer of fireflie.com, my intention is not to advertise here but to show you one example of a site providing this functionality with Python/Django as a back-end.
