If you open the project using eclipse with the same workspace, most of the time when you try to generate signed apk, it will automatically autopopulate with the path where you previously stored. Try doing this. Hope this will help.

There is no fix and no workaround. Even if Google Play would allow you to upload your APK signed with a different key, Android devices would refuse to install your update because of that mismatch. If you are really sure that you cannot find your old keystore then you have to publish your update as a new app with a different package name.
